There are generally three methods for synthesizing Arbidol hydrochloride Cas number 131707-23-8. The first one is to prepare the key intermediate 5-acetyl Ethyloxy-1,2-dimethyl-1H-indole-3-carboxylic acid ethyl ester by the reaction of Nenitzescu, O-acylation and N-methylation with p-benzoquinone and ethyl 3-aminocrotonate, and then the target compound is synthesized by bromination, condensation and Mannich reaction. The method uses the Nenitzescu reaction to synthesize an anthracene ring. The rate is less than 45%, resulting the total yield is less than 25%. Class 1 solvent, 1,2-dichloroethane and carbon tetrachloride is used in the preparation process, which is difficult to use in industrial production because of its toxicity.
Secondly, thiophenol is substituted with ethyl chloroacetoacetate to introduce a phenylthio moiety in the molecule, which is then condensed with methylamine, followed by Nenitzescu reaction with p-benzoquinone, and introduction of dimethylamine by Mannich reaction. Base, after acetylation protection reaction, bromination reaction, then deprotection reaction, and finally salt formation of arbidol hydrochloride.
Thirdly, 1). using p-nitrophenol as a raw material, acetylation protection reaction to obtain p-nitrophenyl acetate;
2) p-nitrophenyl acetate by nitro reduction reaction to obtain 4-acetoxyaniline;
3) 4-acetoxyaniline and ethyl acetoacetate are subjected to an anthracene ring reaction to obtain ethyl 5-acetoxy-2-methylindole-3-carboxylate;
4). The methylation of ethyl 5-acetoxy-2-methylindole-3-carboxylate obtained in the step 3 is methylated to obtain 5-acetoxy-1,2-dimethylhydrazine- 3-carboxylic acid ethyl ester;
5). The 5-acetoxy-1,2-dimethylindole-3-carboxylic acid ethyl ester obtained in the step 4 is subjected to a bromination reaction to obtain 1-methyl-2-(bromomethyl)-5. -acetoxy-6-bromoindole-3-carboxylic acid ethyl ester;
6). The ethyl 1-methyl-2-(bromomethyl)-5-acetoxy-6-bromoindole-3-carboxylate obtained in the step 5 is subjected to thiophenolation and deprotection to obtain 6 -ethyl bromide-5-hydroxy-1-methyl-2-(phenylthiomethyl)indole-3-carboxylate;
7). The 6-bromo-5-hydroxy-1-methyl-2-(phenylthiomethyl)indole-3-carboxylic acid ethyl ester obtained in the step 6 is subjected to Mannich reaction to obtain arbidol, which is salted to obtained arbidol hydrochloride. Cas number 131707-23-8.
