5-fluoro-2-nitroanisole CAS number is 448-19-1, which is the starting material for the synthesis of Osimertinib, 5-fluoro-2-nitroanisole by amine hydrolysis, reduction, nitration, and then The Lewis acid reacts, reacts with N-methyl hydrazine, reductively reduces the nitro group, and amidates to get the product, and finally forms a salt to obtain Osimertinib. 5-fluoro-2-nitroanisole is involved in the amination process: weigh 50g of 5-fluoro-2-nitroanisole into a three-necked flask equipped with a thermometer, add 300ml of DMF, and heat up to 85℃, After stirring for 10 min, 31.4 g of N,N,N'-trimethylethylenediamine was added dropwise, and the mixture was stirred for 4 hours. After the reaction was completed, 600 ml of water was added dropwise. After stirring for 3 hours, a yellow solid was obtained by filtration.
