2-[( Diphenyl methyl)sulfinyl] acetic acid methyl ester is intermediate of modafinil CAS number 63547-25-1. manufacturers automatically withdraw because of too low profits. Some manufacturers cannot compete due to the high cost of their own technical routes. We feel that there is a method that is more suitable for industrial production. His synthesis process is as follows:
 1. In a 1000ml three-necked flask equipped with a stirrer, a thermometer, a condensing device, and an exhaust gas absorption device, add 50g of diphenylmethanol, 200ml of 30% hydrochloric acid, and heat the oil bath to control the internal temperature at 55±5°C and quickly within 10±5min. A mixture of 33 ml of 30% hydrochloric acid and 26.5 g of mercaptoacetic acid was added dropwise. After stirring for about 15 minutes, solids precipitated, and the temperature was raised to 96±3° C., and the reaction was incubated for 2 to 2.5 hours. Toluene 500 ml was added, and the internal temperature was increased by heating. About 75°C, the solid was dissolved with stirring and used for the next reaction.
2. Pour the reaction solution obtained in the first step into a 2L separatory funnel, allow it to stand for about 3-5 minutes, separate the layers, and remove the lower layer of water completely. Transfer the upper toluene layer to the other with stirring, condenser, and thermometer. Tail gas absorption device in a 1L triple reaction flask.
The toluene solution of diphenylmethyl thioacetic acid was heated in an oil bath to 75±5° C., and 70 ml of thionyl chloride and 40 ml of a toluene solution were added dropwise at this temperature. The solution was dropped within 15±5 minutes, and then washed with 10 ml of toluene. The pressure dropping funnel was used for the heat preservation reaction for 2.5 hours, cooled to below 10°C with ice water, and the toluene solution of dibenzylthioacetyl chloride was sealed and stored.
3. In a 2L three-mouth reaction flask with a stirrer and thermometer, add 360ml of ≥28% ammonia, cool to below 0°C in an ice-salt bath, add 430ml of constant water at 0-5°C, and drop at <10°C. The toluene solution of benzhydryl acetyl chloride was added dropwise with vigorous stirring and completed dropwise within 0.8±0.2 h. The reaction was stirred at <10° C. for about 1.0 h, filtered, and the filter cake was washed with purified water to about pH 7. After filtration, the mixture was dried in a baking oven at 60° C. for 6-8 hours to obtain 60.0-65.0 g of Modafinil Carboxylate Methyl Ester .Since 2017, the supply of Modafinil Carboxylate Methyl Ester has been significantly insufficient, and supply and demand have been very tight.
