Rivaroxaban intermediate 4-(4-Aminophenyl)morpholin-3-one CAS number is 438056-69-0, his traditional manufacturing method is the reaction of p-fluoronitrobenzene and 3-morpholinone, and then process catalytic hydrogenation to get 4‐(4‐aminophenyl)‐3-morphinone. The synthesis of this method is difficult, the yield is low, the use of expensive palladium metal is required in the process, the raw material cost is high, and hydrogen is required in the production process, and the relative risk is relatively high.
One method is suitable for the industrial production of 4-(4-aminophenyl)-3-merolinone CAS number 438056-69-0, and 50 g of 3-morpholinone is added to 100 mL of tetrahydrofuran at 10 ℃ and stirred. 24 g of sodium hydrogen was added in portions, and after completion, the reaction was carried out for 30 minutes, and then the temperature was raised to room temperature until the reaction was completed, and the reaction liquid was obtained, which was prepare to use. The reaction solution obtained by the above reaction was added to 67 g of a solution containing 1,4-p-difluorobenzene in tetrahydrofuran at 10℃, and the temperature was naturally raised to room temperature, and the reaction was allowed to keep temperature for 3 hours, and then the temperature was raised to 68℃to reflux till complete. After completion of the reaction, the mixture was cooled to room temperature, and 500 mL of water was added thereto, and the mixture was stirred and filtered, and dried to give 82 g of a white solid, which was 4-(4-Aminophenyl)morpholin-3-one CAS 438056-69-0.
