Product Description

     The ticagrelor intermediate 4,6-Dichloro-2-(propylthio)pyrimidine-5-amine CAS number 145783-15-9, his synthesis process is below:

1. Add 4,6-dihydroxy-2-mercaptopyrimidine to water, and add 10% NaOH and bromopropane in methanol at a temperature of 20-30℃, then raise the temperature to 30-40℃, and adjust the temperature for 11-13 h. After cooling to room temperature, the pH was adjusted to 2-3, stirred, filtered and dried to give 4,6-dihydroxy-2-propylthiopyrimidine.

2. Add glacial acetic acid and fuming nitric acid to the container, cool to low temperature, and then add 4,6-dihydroxy-2-propylthiopyrimidine in batches. The temperature is controlled at -5~10 °C; plus, 0~5 °C After reacting for 3-5 h, after the reaction is completed, it is slowly poured into ice water, stirred and crystallized, filtered, and dried to obtain 4,6-dihydroxy-5-nitro-2-propylthiopyrimidine.

3. Add 4,6-dihydroxy-5-nitro-2-propylthiopyrimidine, POCl3, and triethylamine to the vessel at a controlled temperature of 30-40 ° C; after the addition, the temperature is raised to 110-125 ° C. The reaction is carried out for 4-6h; it is cooled to room temperature, poured into ice water and quenched, stirred with petroleum ether and filtered; the filter cake is rinsed once with petroleum ether, then separated, and the aqueous layer is extracted with petroleum ether, and then the organic layer is combined. Washed with saturated NaHCO3 solution, saturated brine and water, dried over anhydrous magnesium sulfate, filtered, filtered, washed with petroleum ether, and then dried to give 4,6-dichloro-5-nitro-2-propylthio Pyrimidine.

4.
