Baricitinib intermediate 8 CAS number is 1919837-50-5 with the chemical name of 3-Azetidineacetonitrile, 1-(ethylsulfonyl)-3-[4-(4,4,5,5-tetramethyl-1,3,2-dioxaborolan-2-yl)-1H-pyrazol-1-yl]-. In practice, the Baricitinib intermediate 8 often has problems such as difficulty in separation and purification, poor stability, and deterioration. But as long as the method is right, it can also effectively solve these problems. It has been reported in the literature that 3-hydroxyazetidine is protected with ethanesulfonyl group, and then the corresponding intermediate is obtained by oxidation reaction and wittig reaction, and then reacted with pinacol ester of 4-pyrazoleboronic acid to obtain the target compound Baricitinib intermediate 8. The starting material of 3-hydroxyazetidine in the route is relatively expensive and leads to excessive cost, so that it is difficult to be used in industrial production. After verification, we feel that there is a method that is more feasible. The general process is as follows:
(1).1-Amino-3-chloropropyl-2-ol hydrochloride is reacted with ethylsulfonyl chloride under the action of a base to give N-(3-chloro-2-hydroxypropyl)ethanesulfonamide;
(2) N-(3-chloro-2-hydroxypropyl)ethanesulfonamide is subjected to a ring reaction under the action of a base to obtain 1-(ethylsulfonyl)azacridin-3-ol;
(3) 1-(Ethylsulfonyl)azacridin-3-ol is oxidized in a suitable system to give 1-(ethylsulfonyl)azacridin-3-one;
(4). 1-(Ethylsulfonyl) azeridin-3-one is subjected to Wittig reaction with triphenylphosphine acetonitrile, and subjected to a series of simple treatments to obtain 2-[1-(ethylsulfonyl)-3- Azetidinium]acetonitrile;
(5). 2-[1-(ethylsulfonyl)-3-azetidinyl]acetonitrile is deprotected by acid and reacted with neopentyl glycol to give 4-(5,5-dimethyl Base-1,3,2-dioxaborolan-2-yl)-1H-pyrazole
(6). 2-[1-(Ethylsulfonyl)-3-azetidinyl]acetonitrile and 4-(5,5-dimethyl-1,3,2-dioxaboroborane- The 2-based)-1H-pyrazole is subjected to an addition reaction under the action of a catalyst to obtain the target compound Baricitinib intermediate 8 CAS number 1919837-50-5.
