Product Description

Nintedanib intermediate 4 CAS number is 1174335-83-1, the general operation is taking reaction of 2-Oxindole-6-carboxylic acid methyl ester and acid chlorination reagent chloroacetic anhydride to get chloride compound, then the compound and the original three Methyl ester to take condensation to obtain the target compound nintedanib intermediate 4. His specific synthetic experiment process is as follows: 1. In a 3000mL four-necked round-bottomed flask, equipped with mechanical stirring, reflux condenser, thermometer, a round bottom flask was added 1500mL toluene with protection of nitrogen, 240g 6- methoxycarbonyl -2 - oxoindoline, 272g chloroacetic anhydride, open stirring, warmed to 80 ~ 90 ℃, keep warm and stirred for 6 hours, turn off the heating, naturally reduced to 80 ℃, 600ml of cyclohexane was added, stirring was continued to cool down to room temperature , A solid precipitated, suction filtered, the filter cake was washed with ice-ethanol ,vacuum dried to give 233.6g.
2. In a 5000 mL three-necked flask, 195 g of the solid obtained in the first step, 1.8 L of toluene and 257 g of acetic anhydride were added.and heated to reflux. Then, 267 g of trimethyl benzoate was added and refluxed for 4 hours. Heating was stopped and cooled to room temperature , A large number of solid precipitation, the reaction system was added 100ml n-hexane, low temperature crystallization,Suction filtration, dried under vacuum to give a 100gram pale yellow solid, the light yellow solid is the target compound Nintedanib intermediate 4 CAS number 1174335-83-1
